Chemistry Tips: How to Use a Syringe Properly

Are you looking to improve your process with chromatography in the lab?

Many science labs use chromatography to analyze samples. It is a useful way to identify chemicals and solve problems. To use it effectively, you will need to learn how it works. This starts with choosing the right equipment.

That is where a chromatography syringe comes in. These allow you to choose an effective needle and solvent. You can experiment to find the best solution for your needs.

If you want to learn how to use a syringe, keep reading for our chemistry tips.

Use the Proper Size Syringe for the Job

Choose the proper size syringe for the job to help prevent chromatography column damage. Use a 10-microliter syringe for injecting volumes between 5 and 50 microliters. Use a 100-microliter syringe to inject volumes between 50 and 500 microliters.

You can use larger syringes to inject larger volumes, but they may cause damage to the column. Never use a syringe with a needle larger than the inlet or outlet ports on the column.

Fill the Syringe with the Correct Amount of Liquid

When filling a chromatography syringe, it is important to use the correct amount of liquid. Too much or too little liquid can lead to inaccurate results. The amount of liquid needed will depend on the type of chromatography you will use.

A good rule of thumb is to fill a syringe to the point where the liquid just barely covers the tip of the needle. Once you have filled the syringe, you should cap it and store it in a clean, dry area until ready for use.

Be Careful Not to Get Any Air Bubbles in the Syringe

To avoid getting air bubbles in your chromatography syringe, be careful not to touch the plunger to the sides of the barrel. Also, make sure the syringe is in the upright position when drawing liquid into it.

Expel Any Air Bubbles from the Syringe

Before using a syringe for chromatography, it is important to expel any air bubbles that may be present. To do this, slowly push the plunger down until it releases the air.

Then, holding the syringe vertically, tap the side of the barrel to help any remaining air bubbles rise to the top. Finally, slowly push the plunger up until it expels the air bubbles.

Be Sure to Hold the Syringe Securely

Chromatography syringes are delicate instruments that must be handled with care. Always hold the syringe securely, using both hands if necessary, to prevent it from slipping and falling.  It is important to hold the syringe securely to prevent accidents and spills.

Insert the Needle at the Correct Angle

When you are inserting the needle into the septum of the chromatography vial, be sure to do so at a 90-degree angle. This will help to prevent the needle from getting stuck in the septum and will make it easier to remove the needle when you are finished.

Assemble the Syringe and Needle Properly

Before starting chromatography, it is important to assemble the syringe and needle properly. The needle should be firmly attached to the syringe barrel and the plunger should be in the correct position.

The O-ring should be firmly in place and the Luer-Lok fitting should be tight. When inserting the plunger tip into the barrel, be careful not to damage the O-ring. If the O-ring is damaged, it will need to be replaced before use.

Wear Gloves to Protect Your Hands from the Chemicals

In order to prevent any skin reactions, it is recommended that you wear gloves when handling the chemicals used in chromatography. If you do happen to get any of the chemicals on your skin, be sure to wash it off immediately and seek medical attention if you experience any burning or irritation.

Use the Plunger to Draw the Liquid into the Syringe

Before drawing the liquid into the syringe, it is important to ensure that the plunger is in the correct position. The plunger should be pushed all the way to the bottom of the barrel before drawing the liquid into the syringe.

This will help to prevent any air bubbles from being drawn into the syringe. Once the plunger is in the correct position, the syringe can be held pointing downwards and the liquid can be drawn into the barrel.

Inject the Liquid Slowly and Smoothly

When you are injecting the liquid into the chromatography column, it is important to do so slowly and smoothly. This will help to prevent the column from becoming clogged and will also help to prevent the column from becoming damaged. If you are injecting the liquid too quickly, it can cause the column to become clogged, which can lead to the column being damaged.

Withdraw the Needle Slowly

Chromatographic syringes can withdraw liquids from narrow-necked containers such as ampoules, vials, and culture tubes. You insert the needle into the container, and you pull the plunger back to withdraw the desired amount of liquid.

You must withdraw the needle slowly and carefully to prevent liquid from entering the needle hub and contaminating the plunger, wrench, or other syringe parts. Likewise, to prevent liquid from leaking out of the container, you must push the plunger must back slowly and carefully.

Avoid Jerky or Sudden Movements When Handling the Syringe

When learning how to use a syringe properly, it is important to avoid jerky or sudden movements. This can damage the syringe and lead to inaccurate results.

Choose the Right Filter

When using a chromatography syringe, it is also important to choose the right filter. Nylon syringe filters are a good choice because they are durable and can be used with a variety of different solvents.

Wear Gloves to Avoid Introducing Any New Contaminants

In order to maintain the purity of your sample and prevent the introduction of contaminants, it is important to handle your chromatography syringes with gloves. Always put on gloves before touching the syringe or any of its parts, including the needle. Once the syringe is assembled, be careful not to touch the needle or allow it to come into contact with any surfaces. When you are finished using the syringe, dispose of the gloves and wash your hands.

Do Not Touch the inside of the Syringe Barrel with Your Fingers

When handling chromatography syringes, it is important to not touch the inside of the syringe barrel with your fingers. This can contaminate the syringe and lead to poor results.

Label the Syringe with the Name of the Sample and the Date

It is always a good idea to label the syringe with the name of the sample and the date. This will help to ensure that you are using the correct sample and that you are using it within the correct time frame.

Be Sure to Clean All Syringe Parts before Use

Before using a chromatography syringe, it is important to clean all syringe parts. This includes the barrel, plunger, and needle.

To clean the barrel and plunger, disassemble the syringe and soak all parts in a cleaning solution. Be sure to rinse all parts thoroughly with water before reassembling the syringe.

To clean the needle, remove it from the syringe and soak it in a cleaning solution. Be sure to rinse the needle thoroughly with water before reattaching it to the syringe. Any dirt or debris left on the syringe can contaminate the sample.

Allow the Syringe Parts to Air Dry before Reassembling

You should take the chromatography syringe apart and allow the parts to air dry before reassembling. This will prevent the possibility of contamination and cross-contamination of the sample.

Placed the parts on a clean paper towel or piece of aluminum foil. You should leave the parts to air dry for at least 30 minutes.

Dispose of Syringes According to Local, State, and Federal Regulations

It is important to note that you should dispose of syringes according to local, state, and federal regulations. There are a few different ways to dispose of syringes, but the most common method is to place them in a puncture-resistant container.

Once the container is full, you can dispose of it in a biomedical waste disposal facility. It is also important to label the container as hazardous waste.

Never Flush Chromatography Syringes Down the Toilet

When it comes to disposing of chromatography syringes, there are a few things to keep in mind. First and foremost, you should never flush chromatography syringes down the toilet. This can cause serious damage to your plumbing and the environment.

Instead, you should dispose of syringes in special sharps containers that you can find at most pharmacies. Once the syringe is in the container, make sure to seal it tightly so that it can’t be accidentally opened.
